摘要:对纤维素酶法提取长花天门冬多糖的工艺进行优化,并对其抗炎活性进行初步验证。采用纤维素酶辅助提取长花天门冬多糖,通过单因素试验研究纤维素酶添加量、提取时间、提取温度和提取液pH对多糖得率的影响。在单因素试验的基础上,利用Box-Behnken中心组合设计优化长花天门冬多糖的最佳提取工艺。取多糖200mg/kg经口服灌胃给小鼠,观察给药前后咪喹莫特诱导的小鼠皮肤银屑病样炎症,采用苏木精-伊红染色观察其皮肤形态学改变,并通过q-PCR验证皮肤组织中白介素、肿瘤坏死因子等炎症因子的表达差异。结果表明:长花天门冬多糖最佳提取工艺为:纤维素酶添加量1.55%、提取时间51min、提取温度50℃、提取液pH 4.0,在此条件下,多糖得率为10.86%。同时多糖给药在表观及微观结构上能显著改善银屑病症状并下调炎症因子表达量,表明其具有潜在的抗炎活性。

Optimization of Enzymatic Extraction Process for Polysaccharides from Asparagus longiflorus Franch. and Research on Its Anti-psoriasis Effect

Abstract: Cellulase extraction process of polysaccharide from Asparagus longiflorus Franch. was optimized and its anti-inflammatory activity was preliminarily validated. The polysaccharide from Asparagus longiflorus Franch. was extracted by cellulase-assisted method. The effects of cellulase addition amount, extraction time, extraction temperature and pH of the extraction solution on the yield rate of polysaccharide were studied by single factor experiment. On the basis of single factor experiment, the optimal extraction process of polysaccharides from Asparagus longiflorus Franch. was optimized by Box-Behnken central composite design. Extracted polysaccharide was administered to mice at 200 mg/kg orally by gavage, and the psoriasis-like inflammation induced by imiquimod in the skin of mice was observed before and after polysaccharide administration. The morphological changes of the skin were observed using hematoxylin-eosin staining, and the differential expression of inflammatory cytokines were verified such as interleukin and tumor necrosis cytokine in the skin tissue through q-PCR. Results showed: the optimum extraction process of polysaccharides from Asparagus longiflorus Franch. was as follows: addition amount of cellulase 1.55% , extraction time 51 min, extraction temperature 50 ℃, extraction solution pH 4.0, under these conditions, the polysaccharide yield rate was 10.86%. Besides, polysaccharide administration ameliorated morphological and microstructural symptoms of psoriasis and downregulated the expression level of inflammatory cytokines, which indicated its potential anti-inflammatory effect.
